Pool preparation
A total of 28,800 individualized COS clones
(approximately 0.8 GE) for oil palm and coconut, respectively, were
assembled into 30 8-dimensional pools using the
TECAN®GenesisRSP150 and BIOMEK®FX robots (Figure 1).
The pooling strategy resulted in
the unique
presence of each clone in each dimension. DNAs from the 30 pools were
prepared according to the QIAGEN-Tip100 Midi Prep protocol and processed
for AFLP analysis
